Okay, so I'm going to build two tower speakers. I'm going to use standard particle board unless I shouldn't otherwise...

Each Box needs to contain two 10" speakers, a 12" sub, and a tweeter.

Anything you guys have to offer will be helpfull.

Standard particle board will have all kinds of nasty resonances, and give at best mediocre results.

Plywood will resonate less, and more predictably, but hidden knots and hollows in the wood could still hold unwelcome surprises.

Solid wood will be more stable again, but still having grain means that it will have its own resonance, which could colour the sound at certain frequencies.

MDF is nasty nasty stuff, but generally accepted as the best speaker enclosure building material, provided that it is primed and painted once the build is complete. The small particles that comprise the MDF are too small to have any signifigant resonances of their own, but the material isn't airtight, so that's why it needs to be primed and painted. This is what I use for my own speaker cabinets, and I have very picky ears.

Are you making these from a recipe? If so, you want to pay careful attention to what the recipe says. Varying densities and thicknesses can cause unwanted effects. follow it precisely and you should come out with pretty good results. When working with MDF, you want to make sure you pay special attention to your joints and your fit. Measure 5 times and glue once...cause thats pretty much all the chances you will have.

Choose a spot outside to sand these bad boys and make sure you have a fan blowing the particles away from you. The dust they make is like Turbo Pollen and even with a mask its not uncommon to cough up strangley colored globs after sanding....ack ack!

But if you pay close attention to your sanding and fit, you will be much happier with your creations.
